Rybrevant plus chemo shows longest reported survival in EGFR exon 20-mutated lung cancer

Johnson & Johnson (NYSE: JNJ) has reported final overall survival (OS) results from the Phase III PAPILLON trial of Rybrevant (amivantamab-vmjw) plus carboplatin-pemetrexed chemotherapy in previously untreated non-small cell lung cancer (NSCLC) patients with EGFR exon 20 insertion mutations, a historically difficult-to-treat subgroup accounting for roughly 12% of EGFR mutations.

Patients receiving the combination achieved a median OS of 34.3 months versus 27.9 months for chemotherapy alone, a benefit the company said is the longest reported median survival in this population despite 76% of chemotherapy arm patients crossing over to second-line Rybrevant after progression.
